Program 1: Different Cell Culture Techniques

Objective:
To build a strong foundation in mammalian and microbial cell culture, equipping participants with essential theoretical knowledge and practical skills.

1. Aseptic Techniques

3. Subculturing Techniques

  • Trypsinization, cell splitting ratios, and viability assessment

  • Trypan Blue exclusion
  • MTT and other metabolic assays

5. Cryopreservation & Thawing

  • Freezing protocols using DMSO
  • Safe recovery of cell stocks from cryostorage

Program 2: Hematopoietic Stem Cell (HSC) Culture & Analysis

Objective:
To provide comprehensive training in isolating, culturing, analyzing, and characterizing hematopoietic stem cells from blood samples.

Modules:

1. Introduction to Hematopoietic Stem Cells

  • Biology of HSCs: origin, differentiation, and role in hematopoiesis
  • Importance in regenerative medicine and transplantation

2. Identification of Blood Cell Types

  • Use of Romanowsky stains: Leishman, Wright, Giemsa
  • Morphological classification under light microscopy

3. Isolation Techniques

  • ACK lysis for RBC removal
  • Ficoll/Histopaque gradient centrifugation for PBMCs
  • EasySep and Percoll for stem cell enrichment

4. HSC Culture Setup

  • Use of DMEM or RPMI-1640 with 10–20% FBS and antibiotics
  • Seeding in low-adherence plates and maintaining sterility in CO₂ incubators

5. Cell Staining & Microscopic Analysis

  • Trypan Blue viability assay
  • Giemsa, H&E for morphology
  • MTT for cell proliferation/metabolic activity

6. Media Preparation, Subculturing & Cryopreservation

  • Media formulation for HSCs
  • Gentle subculturing of suspension cells
