Program 1: Different Cell Culture Techniques

“From Tissue to Technology — Mastering the Living Laboratory”

Cell culture is more than growing cells — it’s the art of bringing life into a controlled environment.
This hands-on training program takes you through every stage of tissue-based cell growth, from the moment tissue is excised to when cells thrive under the microscope — integrating both classical culture techniques and molecular biology applications.

What You’ll Learn

Introduction to Cell Culture

  • Learn the difference between Primary Cultures, Continuous (Immortalized) Cultures, Adherent Cultures, and Suspension Cultures.
  • Understand the importance of cell culture in diagnostics, drug discovery, and biomedical research.

Instrumentation Demonstration

Hands-on experience with Laminar Flow Hood, CO₂ Incubator, Microscope, Centrifuge, Autoclave, and Water Bath.

Aseptic Techniques & Media Preparation

  • Learn contamination-free handling under biosafety conditions.
  • Prepare DMEM and RPMI-1640, adjust pH, and perform filtration and sterilization steps.

Tissue Processing & Cell Isolation

  • Understand the Modified Tissue Processing Protocol for primary cell culture.
  • Isolate cells from tissue samples and culture them under controlled environmental conditions.

Cell Staining & Microscopy

Perform Trypan Blue Viability Test, Hematoxylin & Eosin (H&E), MTT Assay, and Giemsa Staining to study cell morphology and health.

Cryopreservation & Cell Recovery

  • Resuspension of Cryopreserved Cells: Thawing, gentle centrifugation, and plating.
  • Subculturing & Cryopreservation: Passage cells and preserve them for long-term storage.

Quantitative Techniques

  • Cell Counting & Viability: Using a hemocytometer and Trypan Blue exclusion.

Program 2: Hematopoietic Stem Cell Culture

“Where Blood Becomes Biology — Exploring the Power of Regeneration”

Blood is more than just a fluid — it’s a universe of stem cells, immune cells, and regenerative potential. This specialized module introduces you to hematopoietic stem cells (HSCs) and their vital role in forming blood and immune systems.

What You’ll Learn

Fundamentals of Haematopoiesis

Understand the origin, differentiation, and function of hematopoietic stem cells (HSCs) and how they give rise to all blood cell lineages — red cells, white cells, and platelets.

Identification of Blood Cell Types: Develop the ability to recognize and classify different blood cells under the microscope using Romanowsky stains such as Leishman, Wright, and Giemsa.

Isolation of Hematopoietic Cells: Master multiple isolation techniques, including ACK lysis, Ficoll/Histopaque gradient centrifugation, EasySep, and Percoll density gradient, to separate mononuclear and stem cell populations from blood.

Hematopoietic Stem Cell Culture Setup: Learn how to establish and maintain primary HSC cultures from peripheral blood using DMEM/RPMI-1640 media enriched with FBS and antibiotics, under sterile and controlled CO₂ incubator conditions.

Cell Staining & Microscopic Analysis: Perform essential viability and morphological assessments using Trypan Blue, Hematoxylin & Eosin (H&E), MTT assay, and Giemsa staining to evaluate growth, health, and differentiation.

Media Preparation, Subculturing & Cryopreservation: Gain practical experience in formulating complete media, passaging suspension cells, and preserving hematopoietic cells for long-term storage using DMSO-based cryoprotectants.

Molecular Biology Skills: Isolate genomic DNA from cultured HSCs, run agarose gel electrophoresis, and visualize DNA using a gel documentation system, linking cellular and genetic analyses.

Data Analysis & Scientific Reporting: Analyze, interpret, and present experimental results — from cell counts to DNA profiles — and learn to prepare professional laboratory records, reports, and presentations for academic or research settings.
