Simple staining is a basic staining technique used in microbiology to colorize bacterial cells and make them more visible under a microscope. Here’s a simple staining procedure:

Materials Needed:

  1. Bacterial culture (on a microscope slide)
  2. Bunsen burner or alcohol lamp
  3. Crystal violet (primary stain)
  4. Staining rack or slide holder
  5. Distilled water
  6. Paper towels or bibulous paper
  7. Microscope

Procedure:

  1. Prepare a Bacterial Smear:
    • Place a small drop of water on a clean microscope slide.
    • Aseptically transfer a tiny amount of the bacterial culture to the water on the slide, spreading it to create a thin film. Allow it to air dry.
  2. Heat Fixation:
    • Pass the slide, bacterial side up, through the Bunsen burner flame or over an alcohol lamp several times (do not overheat, as it can distort the cells). This heat fixation process kills the bacteria, adheres them to the slide, and coagulates cellular proteins to make them more stain-absorbent.
  3. Staining:
    • Apply a few drops of crystal violet (primary stain) to completely cover the bacterial smear on the slide. Let it sit for about 1 minute.
  4. Rinsing:
    • Hold the slide at an angle and rinse gently with distilled water to remove excess stain.
  5. Blot Dry:
    • Blot the slide gently with a piece of bibulous paper or a paper towel to remove excess water, being careful not to disturb the bacterial smear.
  6. Microscopic Examination:
    • Place the slide on the microscope stage and focus the microscope on the stained bacterial cells.
    • Observe and study the stained cells under different magnifications to understand their morphology and arrangement.
  7. Cleaning:
    • Clean the slide with water and a gentle detergent after use, and store it properly.

This simple staining procedure helps to highlight the overall morphology, size, and arrangement of bacterial cells. It is a valuable technique for preliminary microscopic examination in microbiology.
